Description |
Ectopic overexpression of FaNYE1 in Arabidopsis results in yellowing phenotypes. A fragment containing the FaNYE1 ORF was amplified by PCR using two oligonucleotides: 5′-AGGGAGCTCGCAGAGAGCCTGATC-3′ and 5′-GCTGG ATCCAGGACCCGTGTACTTG-3′. The PCR products were cloned into pMD19-T vector and sequenced. After digestion, the released fragments were subcloned into SacI and BamHI site of P AtNYE1 -pCHF3.For overexpression of FaNYE1, the DNA fragment containing the ORF of FaNYE1 was digested from P AtNYE1 -pCHF3 and was subcloned into the SacI and BamHI site of pCHF3. Background of Transgenic lines is Col-0. |

FaNYE1, an orthologue of AtNYE1, was further identified from a major type of cool-season turf grass, tall fescue (Festuca arundinacea Schreb.), by RACE-PCR. FaNYE1 encoded an AtNYE1-like chloroplast protein. FaNYE1 could be strongly induced by dark treatment and natural senescence. Constitutive overexpression of FaNYE1 in Arabidopsis resulted in the whole spectrum of leaf yellowing phenotypes, the severity of which correlates with its transcript level. |
