The nucleic acid was extracted from the 140 ul concentrated sewage. Reverse transcription (RT)-PCR was performed by using random primers. The cDNA was then used for two rounds of PCR amplification with specific primer sets. The positive products (~354 nucleotides) were subjected to Illumina MiSeq sequencing (PE250) and around 1,000,000 reads were generated for each PCR product. |
