The chloroplasts of H. cordata were initially enriched from 5 g fresh leaves. Then, the cp DNA was isolated from the chloroplasts using the DNeasy Plant Mini Kit (Qiagen, USA) according to the manufacturer’s instructions. After assessing the quality and quantity of the cp DNA, one large DNA library with 20 kb insertion for PacBio Sequel (PacBio, USA) platform sequencing and one small DNA library with 450 bp insertion for Illumina HiSeq X Ten (Illumina, USA) sequencing were constructed based on the manufacturer’s instructions. |
