5 μg of genomic DNA was fragmented by sonication to a mean size of approximately 200-400 bp, followed by end repair, A-base tailing and adapter ligation using the Acegen DNA Library Prep Kit (Acegen, Cat. No. AG0810) according to the manufacturer’s protocol. The adaptor-ligated DNA was immunoprecipitated by the anti-6mA antibody (Synaptic Systems, catalog number 202011). The captured DNA was then purified and amplified by 14-17 cycles of PCR using Illumina 8 bp dual index primers. Simultaneously, the input library was obtained from 5-10 ng of the ligated DNA before immunoprecipitation. The constructed libraries were then analyzed by Agilent 2100 Bioanalyzer and finally sequenced on Illumina platforms using a 150×2 paired-end sequencing protocol. |
