C57BL/6N female mice were induced to superovulation by injections of 5 IU of pregnant mare’s serum gonadotrophin(Ningbo Sansheng Pharmaceutical Co, Ltd) and 5 IU of human chorionic gonadotrophin(Ningbo Sansheng Pharmaceutical Co, Ltd) 44-48 hours apart and mated to PWK males. Embryos were flushed out from uterus at E4.5 and cultured 4 days in CMRL 1066 supplemented with 1mM glutamine, 1mM sodium pyruvate and 20% fetal bovine serum. After trypsin and pancreatic enzyme digestion followed by mechanical dissection to separate epiblast and visceral endoderm, embryonic cells were washed with PBS and incubated in lysis buffer.Hi-C libraries were prepared using Hi-C protocal developed by Rao et al. with slightly modification. |
