A total of seven tissues representing different developmental stages of Oryza granulate were sampled to generate the RNA-Seq data needed for subsequent genome annotation. These tissues included panicles at three different stages of flower development, flag leaves, and stems and the shoots and roots of three-leaf seedlings. Because of the low germination rate of O. granulata, seedlings were germinated from seeds harvested from multiple plants, while the remaining tissues were sampled from the individual used for genome sequencing. All collected samples were quickly frozen in liquid nitrogen. RNA was individually extracted from each tissue using TRI reagent according to the instructions provided with the reagents. |
