1 mg total genomic DNA was randomly broken into fragments at the size of around 350 bp by the method of ultrasonication. The library was sequenced by Illumina HiSeq 2500 after passing quality inspection. Variation detection by high-throughput sequencing for two parental individuals (B73, Abe2) and two offspring mixed pools (S-pool, T-pool). Based on the detected SNPs, a Bulked Segregant Analysis (BSA) method was used to map QTLs and detect relevant gene loci. |
