The high-molecular-weight DNA was extracted from in vitro seedlings using the Qiagen MagAttract HMW Kit (PN-67653), following the Chromium Genome Protocol. The HMW DNA (~50kb in length) was loaded onto a Chromium Controller chip with 10X Chromium reagents and gel beads for the library construction. The resulting library was sequenced on Illumina HiSeq X Ten sequencer to produce PE150 reads.
