HD701 was mixed with Hapmap Normal cell line NA18536.Diluted DNA mixtures were further sheared using a focused-ultrasonicator with a target size around 170 bp to mimic the size distribution of cfDNA. 33 ng sheared reference sample was used for end repair and A-tailing, followed by ligation of SinoDuplex adapters and amplification using KAPA HiFi HotStart ReadyMix with unique dual indexes.Targeted capture was performed with customized panel according to IDT xGen Hybridization and Wash Kit protocol. |
