Feces samples about 0.5 grams each were collected from a total of four healthy Chinese. Total DNAs were extracted using the Stool DNA kit D4015 (Omega, USA) and then detected the integrity and concentration respectively by 1% agarose gel electrophoresis and the super-micro spectrophotometer (Thermo NanoDrop2000). The DNAs were then divided into two parts and submitted to NGS and TGS sequencing. For the NGS sequencing, the DNAs were randomly interrupted for constructing the 500bp sequencing library (NEB, USA), then conducted Hiseq4000 pair-end sequencing. |
