Feces samples about 0.5 grams each were collected from a total of four healthy Chinese. Total DNAs were extracted using the Stool DNA kit D4015 (Omega, USA) and then detected the integrity and concentration respectively by 1% agarose gel electrophoresis and the super-micro spectrophotometer (Thermo NanoDrop2000). The DNAs were then divided into two parts and submitted to NGS and TGS sequencing. TGS sequencing was performed by the standard SMRT sequencing library (SMRTbell Template Prep Kit 1.0) for PacBio Sequel sequencing. |
