Total RNA was extracted from the fully unfolded leaf blades located at the same position on the main stems of 40-day-old plants using RNeasy Plant Mini Kit (Qiagen), according to the manufacturer’s instructions. The concentration and quality of total RNA were evaluated using a NanoDrop spectrophotometer and Agilent 2100 Bioanalyzer, respectively. Libraries for RNA-Seq analysis were constructed from 3 μg of total RNA per sample using NEBNext®Ultra™ RNA Library Prep Kit for Illumina (NEB), according to the manufacturer’s recommendations. Each sample was then labeled with a unique index code. Cluster generation was performed using TruSeq PE Cluster Kit v4-cBot-HS (Illumina). After cluster generation, the libraries were sequenced using the Illumina HiSeq 2500 platform (Berry Genomics) to generate paired-end reads. |
