The Hi-C experiment was performed follow the in-situ Hi-C protocol. Briefly, cells were lysis and chromatin was digested by MobI, then biotin fill-in and Hi-C ligation. After ligation, DNA was purified and sonicated to 200-500bp for library construction follow the . DNA was end-repaired, dATP attached and adapter ligated follow the in-situ Hi-C protocl. After adapter ligation DNA was PCR amplified and library fragments between 200-500bp were purified for sequencing. The purified library was sequenced on NovaSeq at BerryGenomics Co., LTD. |
