Total RNA was extracted from the two-week-old seedlings of Col. 5 ?g of total RNA was treated with RQ1 DNase (Promega, M6101) and was partially digested by RNase T1 (Thermo, EN0541), an endoribonuclease that specifically digests single-stranded RNA at the 3?-side of the G residue. Poly(A)-containing RNA fragments were purified with oligo(dT)-conjugated magnetic beads (Invitrogen, 61005) and was ligated to the 3?-RNA adaptor. The cDNA was synthesized using ScriptSeq™ v2 RNA-Seq Library Preparation Kit (Illumina, SSV21124). PCR products of 200-500 bp were purified and were applied to HiSeq X ten system for 150-nt pair-end sequencing. |
