The poly (A) mRNA was isolated by magnetic oligo (dT) beads, fragmentation buffer was added at appropriate temperatures to breaking the mRNA into short fragments. First-strand cDNA was synthesized by the interrupted mRNA as a template using random hexamer-primer. The second-strand cDNA was synthesized using buffer, dNTPs, RNaseH and DNA polymerase I. Short fragments were purified using Min Elute PCR Purification Kit (Qiagen, China), sticky-end repair and tailing A. Then, the short fragments were ligated to sequencing adapters, selected the suitable size of the fragment by the 1% agarose gel electrophoresis and performed the PCR amplification. Finally, the library was sequenced using Illumina HiSeqTM 2000 platform |
