~0.1 g of sample was used for ChIP-seq assay. Samples were fixed with formaldehyde under vacuum and stored at –80 °C until use. Chromatins were isolated with high effective method and fragmented into 200–600 bp with Bioruptor plus. ChIP assay was performed with H3K27me3 (ABconal, A2363). After reverse–cross–linking the protein-DNA complexes, immunoprecipitated DNA was purified by phenol:chloroform:isoamyl alcohol extraction. ChIP DNA was used for library preparation per instructions (NEBNext Ultra II DNA Library Prep Kit, New England Biolabs), and libraries were sequenced on the Illumina HiSeq X Ten platform. |
