Total RNA was extracted with an RNeasy plant mini kit from the whole seedlings of wild type and miR408 transgenic line grown on MS for 10 days under continuous light in a growth chamber. The mRNA fraction was extracted from total RNA using Dynabeads oligo(dT)25. Double-strand cDNA were generated using Superscript II and random hexamer primers and fragmented by nebulization, and used for mRNA library construction. |
