Zhe-Maidong (Ophiopogon japonicus (L.f.) Ker-Gawl), a traditional medicinal herb in the Liliaceae family, which has shown great pharmacological effects on immunity adjustment and cardiovascular disease, the planting period is generally three years. In this study, three different growth stage of Zhe-Maidong were used to RNA-seq, and total16.4G raw data were obtained. After filtering and assembling, 96,738 unigenes were finally generated with an average length of 605.3 bp. Total 77,300 unigenes were got annotation information for five databases, including NT, NR, Swiss-Prot, KEGG and GO. In addition, the mechanism of flavonoids, saponins and polysaccharides formation and accumulation at different stages of tuber development were also characterized. From first year to third year, the content of flavonoids, saponins and polysaccharides were all increasing, whereas the expression level of related genes were decreasing in pathway of flavonoids and saponins, and first increasing and then decreasing in the pathway of polysaccharides. In conclusion, the results of this study provide the most comprehensive expressed sequence resources for Zhe-Maidong, which will enlarge the available O. japonicas gene library and facilitate further genome-wide research and analyses in this species. |
