Two-week-old MM and SlymiR482e-3p-knock-out (SlymiR482e-3p-KO) seedlings were infected with either mock (water) or Fol for 24 hours. Roots were collected from three biological replicates (20 seedlings each). Total RNA was extracted using TRIzol Reagent (15596026, Life Technologies, CA, USA), and mRNA was purified with magnetic beads with Oligo (dT). For each library, 2 μg total mRNA was applied to generate the Illumina library according to the manufacturer instruction. The libraries were subsequently sequenced using Illumina HiSeqTM 2000 (Biomarker Technologies). |
