Metabolic reprogramming is considered as a hallmark of cancer and metabolic manipulation such as targeting fatty acid (FA) structural modification will be a promising therapeutic approach in cancer treatment. The enzyme fatty acid 2-hydroxylase (FA2H) specifically introduces a chiral (R)-hydroxyl group at the second carbon of FAs, leading to the generation of (R)-2-hydroxylated FAs ((R)-2-OHFAs). The potential anti-tumor activity of FA2H and its product (R)-2-OHFAs has been suggested in several cancer types. In the present study, we aimed to investigate the potential role of FA2H in the development and progression of colorectal cancer (CRC). mRNA profiles of wild type (WT) and FA2H over expressed LOVO cells were generated by deep sequencing, in triplicate, using Illumina sequencing platform.Sequenced reads were trimmed for adaptor sequence, and masked for low-complexity or low-quality sequence, then mapped to hg19 whole genome using tophat2 v2.1.1 with parameters --no-novel-juncs -p 10. GFOLD v1.1.4 was utilized to count reads to generate gene-level counts according to a protocol from Feng et al., Bioinformatics, 2012. Finally, limma pipeline was used to identify the differentially expressed genes. The RNA-Seq data and our low-throughput experiments define FA2H and its enzymatic product (R)-2-OHPA as novel regulators of Hippo-YAP pathway in CRC cell growth and metastasis, and suggest that (R)-2-OHFAs could be candidates of novel drugs in CRC treatment. |
