Description The CRISPR-Cas9 system is a highly efficient genome editing tool widely used in model and non-model animals with promising potential in human gene therapy. Although with much improvement, the off-target effect of CRISPR-Cas9 remains a concern. Non-human primates (NHPs) share close genetic, immunological, physiological and neural similarity with humans, making them the ideal preclinical models for developing Cas9-based therapies. However, no robust off-target assessment has been conducted in NHPs. Here we performed trio sequencing of five Cas9-treated monkeys and their wild type parents to detect off-target effect of CRISPR-Cas9 system.
