Description We used the siRNA and in situ Hi-C techniques together to validate the effect of Znf143 on chromatin loops. The Znf143 was silenced in HEK293T cells by using siRNA technique, and the in situ Hi-C were performed on both negative control and Znf143-silenced HEK293T cells. The results show that chromatin loops can be both lost and gained after knocking down Znf143 expression level, but the Znf143 silence mainly destabilizes or eliminates chromatin loops.
