Title Unveiling the complexity of the pig transcriptome by single-molecule long-read sequencing

Description Pig is an major genetic model for elucidating transcriptional networks. Uncertainties about the complete structure of mRNA transcripts limit the progress of research in this system. Here, using the single-molecule sequencing technology for analysis the complexity of porcine transcriptomes, we obtained porcine reference annotation from 38 tissues. In parallel, messenger RNA of 8 tissues from the 38 tissues was sequenced on the Illumina PE125 platform to quantify gene or isoform expression. Here we combined NGS (short-read next-generation sequencing technology) and SMRT sequencing to generate a more complete full-lenth porcine transcriptome. Accordingly, this study highlights the splice isoforms and transcriptome diversity and dynamics, provides a valuable resource for further investigation of genome annotation, enhances our understanding of the porcine transcriptome.
